Postdoctoral position in Developmental Neurobiology

A postdoctoral position is available from January 2021 in the “Development and Function of Brain Circuits” team led by M. Studer. We aim to understand how rare genetic variants of key master genes of brain development and circuit assembly can cause neurodevelopmental diseases (NDDs). The project will determine the impact of missense variants identified in the NR2F1 gene on the heterogeneity of neurodevelopmental defects described in affected patients. The postdoc will study genotype/phenotype correlations by using CRISPR/Cas9 gene editing in vivo and decipher the
pathological contribution of representative variants in the mouse system. We expect that the results of this work using experimental systems as well as the identification of Nr2f1 direct targets and interacting partners will help in understanding the phenotypic heterogeneity of patients and unveil pathophysiological mechanisms common to multiple NDDs.

The team is located in the heart of Nice and is part of the Institute of Biology Valrose (27 teams; 300 people; 25 nationalities), an international research centre that brings together high-profile teams with complementary areas of expertise and with a common interest in translating basic research into knowledge for the clinic. Qualification and experience

We are seeking highly motivated young candidates holding a PhD for less than 2 years. Preference will be given to applicants with a background in mouse genetics, cellular and molecular biology, confocal imaging and genome wide approaches. The position is opened for one year renewable up to 3 years (including medical benefits) starting as early as January 2021. Fluency in English is mandatory but ability to speak French is not required.

POST-DOC POSITION IN CELL AND DEVELOPMENTAL BIOLOGY.

Position available (starting oct./dec. 2018) in Pascal Thérond laboratory to functionally characterize Hedgehog secretion and spreading in Drosophila. Hedgehog proteins are key signaling mediators that govern a wide range of processes during embryonic development and adult tissue homeostasis from Drosophila to human. Hedgehogs are the only metazoan proteins known to possess a covalently-linked cholesterol moiety. Our laboratory is interested in how Hedgehog proteins traffic in the producing tissue and exert their function in the receiving tissue.
We have shown that the Endosomal Sorting Complex Required for Transport (ESCRT) promotes Hedgehog proteins loading on exo-vesicles to exert their effect at long distances. The post-doctoral project aims to gain further insight into the trafficking, vesicular secretion and the extracellular spread of Hedgehog proteins through tissues using cell biology and genetic technics. Moreover, in vivo imaging and single molecule tracking (in collaboration with computational science lab) has been developped on our tissue models and will be further used to investigate the dynamics of Hedgehog release and spreading.

Interested candidates should have strong knowledge of, and experience in fly genetics, cell biology and optic microscopy (confocal/spinning disc). The position is funded for 3 years in duration. Candidates must have a Ph.D. degree, and can be nationals of any country.

Projet

Dans un partenariat académiques/industriels, le projet financé par la Banque Publique d’Investissement a pour objectif le développement d’une nouvelle génération de ciments phosphocalciques élastiques et résistants pour le traitement des fractures vertébrales.

Programme de travail et compétences requises

Le programme de travail repose sur l’étude des interactions entre les ciments phosphocalciques développés et les cellules osseuses (ostéoblastes, ostéoclastes), grâce à la mise en oeuvre des techniques suivantes :

• Préparation et mise en forme des ciments pour les tests in vitro ;
• Culture cellulaire et mesures de la viabilité/prolifération ;
• Transcriptomique (RT-qPCR) et protéomique (ELISA multiplex)
• Immunohistochimie

La personne sera aussi impliquée dans l’analyse d’implants en site osseux réalisés par nos collaborateurs responsables de l’expérimentation in vivo, grâce à une technique de microscopie quantitative (SHG, Second Harmonic Generation)
Sont attendus : rigueur et réactivité dans la conduite des expériences et dans la mise en forme des résultats ; aptitudes à interagir au sein d’un réseau impliquant 3 laboratoires académiques et 2 industriels

Regulation of neuronal RNA granules and function in long-term memory

A three-year post-doctoral position is available in the group of Florence Besse

Project description

Long-term molecular and structural changes occurring at synapses in response to neuronal activation provide the mechanistic bases for the establishment and retention of memories, and are altered in both developmental and age-related memory disorders. Such changes rely on local experience-induced translation of quiescent mRNAs packaged together with regulatory proteins into neuronal RNA granules transported and stored at synapses. How RNA granules are remodeled in response to neuronal activity to relieve translation repression of mRNAs is unclear. Furthermore, the importance of such a remodeling in the establishment of long-term memories remains to be demonstrated in vivo.
The successful candidate will address these questions in Drosophila, by dissecting the cellular and molecular mechanisms underlying the remodeling of conserved neuronal RNP granules in response to activity. She/he will also participate in characterizing how such a remodeling impacts on the establishment of long-term memory traces assessed via learning and memory behavior assays (in collaboration with K. Keleman’s lab).

Qualifications and Experience

Candidates should have a PhD in cell biology, molecular biology, genetics, or neurobiology.
Experience with fly genetics, behavior assays and/or transcriptomics will be considered advantageous.
English is the working language in the lab and in the Institute.

PhD Fellowship in Developmental Biology

Maternal factors regulating Nodal expression in the sea urchin embryo:
a mechanistic and evolutionary approach

A three-year PhD position supported by the French Foundation for Medical Research (FRM) is available starting September 1st 2018 to work in the group Gene regulatory networks, axis specification and morphogenesis of the sea urchin embryo at the Institute of Biology Valrose. The area of research will concern the analysis of Dorsal-ventral patterning in the sea urchin embryo through the analysis of the function and evolutionary conservation of several maternal factors that play critical roles in the regulation of nodal expression and/or Nodal activity. Our laboratory has recently identified several maternal factors that play crucial roles in the regulation of nodal expression including a maternal TGF beta called Panda, a BMP type I receptor called Alk1/2, a maternal transcription factor of the Ets family called Yan as well a as novel secreted protein of unknown function. Although these factors are each crucially required to spatially restrict nodal expression, their mechanism of action is not understood. Furthermore, it is not known if these genes are required to restrict nodal expression only in the Mediterranean sea urchin Paracentrotus lividus or if their function is conserved in other species.
The project will specifically address these questions by elucidating how these factors work and determining if their function is conserved in different species.

en oncoimmunologie fondamentale et translationnelle des cancers des VADS

Le Centre Antoine Lacassagne (Nice) et les équipes du Dr Ellen Van Obberghen (iBV, Nice) et du Dr Fabienne Anjuère (IPMC, Valbonne) ont mis en place un consortium afin de potentialiser des travaux collaboratifs en oncoimmunologie fondamentale et translationnelle des cancers des voies aéro-digestives supérieures (VADS). Ce consortium s’intéresse à la compréhension des interactions entre le stroma tumoral et les cellules immunitaires dans les cancers des VADS afin d’identifier de nouveaux marqueurs pronostiques et de nouvelles approches d’intervention thérapeutique.

Dans ce cadre, le consortium recrute un personnel technique, niveau ingénieur ou assistant-ingénieur, en CDD pour une période de 12 mois renouvelables. Le candidat sera chargé de la gestion de biopsies humaines de cancers des VADS dans le cadre de recherches fondamentales et d’essais cliniques d’immunothérapie, de l‘analyse de la signature immunitaire et stromale de ces biopsies humaines par des approches complémentaires d’imagerie en fond clair et de fluorescence et de cytométrie de flux. Ces analyses multiparamétriques requièrent de plus des compétences en informatique et en gestion et archivage de données.

Profil recherché

  •  Expérience en histologie (coupes de biopsies incluses en paraffine et cryocongelées, coloration de lames, marquages en immunohistochimie et en immunofluorescence)
  •  Expérience en cytométrie de flux multiparamétrique et connaissances en immunologie également souhaitées.
  •  Capacité à développer et rédiger des protocoles standardisés
  •  Qualités d’organisation, rigueur et esprit d’équipe indispensables
  •  Disponibilité et flexibilité d’horaires liée au travail sur biopsies humaines fraiches à processer extemporanément

Lieu d’activité: Le site principal de l’activité sera basé à l’IBV, Faculté des Science, Nice, et requerra des missions régulières sur les sites de recherché du Centre Antoine Lacassagne (Centre de Ressources Biologiques), Nice et de l’IPMC, Valbonne.
