"PEPS" - Protein Expression & Purification System

 

Presentation

As of 2008 the « PEPS », dedicated to the expression and purification of proteins, has been made available to all members of the IBV. This platform aims to centralize all requests for expression and purification of proteins, from cell preparation (transfected for overexpression of proteins) up to the pure protein. The platform is located on the 4th floor of Bâtiment Sciences Naturelles.


Equipment

1 Fast Performance Liquid Chromatography (FPLC)
 
A high-pressure computer assisted chromatograph (AKTA FPLC Pharmacia) allows purification of proteins and peptides with instantaneous data acquisition of the absorbance spectra at 280 nm. The advantage of the system: High resolution and quick, with the possibility of working in an Analytical or Preparative mode.
 
A range of columns for purification (depending on the separation parameters)
 
   
2 filtration columns
    (1 Superdex 200 et 1 Sephacryl S300).
1 hydrophobic interaction column
2 ion exchange columns (1 anionic, 1 cationic).
1 Heparin affinity column
    (for purification of growth factors in particular).
1 GSTrap column (5ml)
1 Hitrap Proteine G column (5ml)
41 affinity columns (made in house) for the purification
    of polyclonal antibodies from mouse or rabbit sera.
   
1 lyophiliser (Kinetic Flexi dry) with a capacity of 1 litre/24h.
In addition to its own equipment the platform has access to complementary equipment of other laboratories.
3 centrifuges
3 shaking incubators
1 spectrophotometer
1 FRENCH press (Tour Pasteur)

 

Development of the platform

The purchase of new columns and chromatographic equipment will allow further development of more complex protocols in order to satisfy an even larger number of users.
