FACSAria Technical Information

The platform is located on the 4th floor of Bâtiment Sciences Naturelles.
The FACSAria is a fixed-alignment benchtop high-speed cell sorter. It is equipped with three lasers allowing multicolor analysis of up to 11 parameters:

   
405 nm excitation
  • 450/50: Pacific Blue, DAPI
  • 530/30: AmCyan
   
488 nm excitation
  • 530/30: FITC, Alexa Fluor 488
  • 585/42: PE
  • 616/23: PE-Texas Red
  • 695/40: PerCP-Cy5.5, PI
  • 780/60: PE-Cy7
   
633 nm excitation
  • 660/20: APC, Alexa Fluor 647
  • 780/60: APC-Cy7, APC-H7
   
Forward scatter detector
  Diode and Photo Multiplier Tube combination allows for analysis of traditional flow cytometry cell samples, but also provides enhanced dynamic range to more reliably detect and analyze bacteria and other small organisms.
   
Side scatter detector
  PMT with 488/10 filter

 
Other features

  • Interchangeable 70 and 100-µm orifice sort nozzles which permit sorting of a variety of cells types - bacteria up to large, fragile mammalian cell lines - at a wide range of pressure settings

 

  • Aseptic Sorting
    An automated cleaning protocol disinfects the fluidics system with 10% bleach and 70% ethanol when aseptic sort conditions are required. Both sample input and sort collection areas are closed aseptic chambers.

 

  • Sample Input formats
    Filtered single-cells suspensions can be delivered for sorting in 1.5 mL, 5 mL (12 x 75 mm ), or 15 mL polystyrene or polypropylene tubes. Samples can also be agitated at various speeds to keep cells suspended during sorts.

 

  •  A variety of Sort Collection Devices
     
    • Tubes
    • 2-way sorting into 5 mL (12 x 75 mm ) or 15 mL tubes
    • 4-way sorting into 1.5 mL microtubes or 5 mL (12 x 75 mm ) tubes
    • Plates and slides through the Automated Cell Deposition Unit (ACDU)
    • Standard-size glass slides or 6, 24, 48, 96, and 384-well plates

 

  • Temperature Control
    Both sample input and sort collection chambers can be maintained at temperatures ranging from 4 to 42°C
